Human Cellular and Molecular Neuropharmacology

Group
The laboratory has developed in-vitro models to study the mechanism of action of psychoactive drugs in human and rodent CNS neurons. Presently, we are involved in projects where dopaminergic and cortical neurons differentiated from iPSC donated by patients with neurodegenerative and psychiatric disorders are used to identify disrupted molecular pathways and assess their response to various pharmacological agents, so to identify the right treatment for the right patient. One of the projects addresses clinically and immunologically characterized patients with Treatment Resistant Depression. Another project addresses the characterization of molecular risk factors and potential novel targets relevant in parkinsonian disorders including GBA-Related Parkinson’s Disease and Multiple System Atrophy. The laboratory is also engaged in assessing novel devices and technologies aimed to improve the molecular and phenotypic profile of iPSC-derived neurons. Il laboratorio ha sviluppato modelli neuronali in vitro per studiare il meccanismo d’azione di farmaci e di agenti candidati attivi sul sistema nervoso centrale utilizzando cellule di derivazione umana e murina. Attualmente siamo coinvolti in progetti di ricerca finalizzati allo studio di malattie neurodegenerative e psichiatriche utilizzando neuroni e glia derivati da iPSC donate dai pazienti. Questi studi hanno una finalità di Medicina Traslazionale: lo scopo è di caratterizzare substrati molecolari e fenotipi cellulari alterati associati alla malattia e la loro risposta ad agenti farmacologici con potenziale terapeutico. Un progetto di priorità è dedicato allo studio di soggetti con Depressione Maggiore Resistente alla terapia caratterizzati clinicamente e per il loro profilo immunologico. Un altro progetto si focalizza sullo studio degli aspetti molecolari rilevanti nei parkinsonismi, in particolare la Malattia di Parkinson associata a mutazione del gene GBA e l’Atrofia Multisistemica (MSA). Un terzo progetto è mirato all’adeguamento tecnologico dei modelli da noi utilizzati in collaborazione con centri di ricerca di bioingegneria.
